چکیده

aim :  to evaluate the association of dnmt1 and mgmt amino acid substitution polymorphisms and colorectal cancer in iranian population. background : the mgmt and dnmt1 are two important methyltransferase enzymes. amino acid substitution polymorphisms in mgmt and dnmt1 genes may be associated with the genetic susceptibility to sporadic colorectal cancer. patients and methods : we assessed eight non-synonymous polymorphisms of these two genes by pcr/pyrosequencing. our population consisted of 208 individuals with sporadic colorectal cancer and 213 controls. allele frequencies and genotypes were compared between the cases and controls. results : odds ratios were calculated and there was no association between dnmt1 and colorectal cancer. however, there was a significant association between two polymorphisms with sporadic colorectal cancer; arg128gln (or 5.53, 95%ci 2.58-7.16) and gly160arg (or 3.04, 95%ci=1.48-6.31) of mgmt gene. conclusion : this finding could be an accurate indicator of high occurrence of colorectal cancer in iranian patients.

Effects of amino acid substitution polymorphisms of two DNA methyltransferases on susceptibility to sporadic colorectal cancer.

BACKGROUND AND AIM The present study was designed to consider whether amino acid substitution polymorphisms in O6-methylguanine-DNA methyltransferase (MGMT) and DNA methyl transferase 1 (DNMT1) genes may be associated with the genetic susceptibility to sporadic colorectal cancer. PATIENTS AND METHODS We assessed eight non-synonymous polymorphisms of these two genes by PCR/pyrosequencing. Our ...

بررسی نقش پلی مورفیسم‌های ژن O6 – متیل گوانین متیل ترانسفراز در سرطان کولورکتال

Abstract Background: Sporadic colorectal cancer is the fourth most common cancer in Iran. The DNA repair protein O6-methylguanine-DNA methyltransferase (MGMT) is involved in the cellular defense against alkylating agents. Genetic alterations in the MGMT gene may impair the protein’s ability to remove alkyl groups from the O6-position of guanine, thereby raising the mutation rate and increasi...

Amino acid substitution polymorphisms of the DNA repair gene MGMT and the susceptibility to cervical carcinoma.

In the current study, we examined the association between polymorphisms in the O(6)-methylguanine-DNA methyltransferase gene (MGMT) and the risk for cervical carcinoma. We prospectively selected 1012 patients, including 539 with carcinoma and 473 with cervical intra-epithelial neoplasia and 800 healthy women from five hospitals in Zhejiang Province, China. Three single-nucleotide polymorphisms ...

Association between polymorphisms of TAS2R16 and susceptibility to colorectal cancer

BACKGROUND Genetics plays an important role in the susceptibility to sporadic colorectal cancer (CRC). In the last 10 years genome-wide association studies (GWAS) have identified over 40 independent low penetrance polymorphic variants. However, these loci only explain around 1‑4% of CRC heritability, highlighting the dire need of identifying novel risk loci. In this study, we focused our attent...

RAD51 gene polymorphisms and sporadic colorectal cancer risk in Poland.

BACKGROUND DNA repair processes play an important role in protection against carcinogenic factors. Mutations in DNA repair genes, which code proteins engaged in repair processes, may lead to carcinogenesis and among others also to colorectal cancer (CRC) development. The genetic variability in RAD51 may contribute to the appearance and progression of various cancers including CRC. The aim of th...
